Thomas Luke Macfarlane was born in London, Ontario, on January 19, 1980. His father, Thomas, worked as the Director of Student Health Services at the University of Western Ontario, while his mother, Penny, worked as a mental health nurse in a London hospital. Ruth, Macfarlane’s twin sister, and Rebecca, Macfarlane’s older sister, both attended London Central Secondary School.

Macfarlane attended the Lester B. Pearson School for the Arts and then moved on to Juilliard in New York City to study acting.
